The Yeaman group is interested in identifying molecular mechanisms responsible for biogenesis of epithelial cell polarity (publications in Cell, Molecular Cell, Nature Cell Biology, J. Cell Biol., etc). We study details of fundamental cellular processes, such as intercellular adhesion, cytoskeletal remodeling and membrane trafficking, that are at the foundation of tissue development and organogenesis. Our focus is on the Exocyst, a multi-functional, allosterically regulated scaffold, and Ral GTPases, which regulate Exocyst functions by facilitating association or disassociation with additional proteins. We utilize CRISPR/Cas9 gene editing, along with mouse models, 2D and 3D cell culture models, cutting edge cell imaging technologies and biochemical assays to understand how Exocyst protein assemblies function during cell growth and differentiation, motility and cell division. We are performing focused structure-function studies in efforts to understand how specific mutations in genes encoding Exocyst and Ral proteins contribute to human diseases including cancers, diabetes and ciliopathies.
